    Alomone Labs 4-ONE activates TRPA1 channels expressed in HEK-293 cells.
    Dose response curve of the activation of human TRPA1 channels by increasing concentrations of 4-ONE (#O-145). Cells were loaded with Calcium 5 dye. Changes in intracellular Ca2+ following 4-ONE application were detected as changes in relative fluorescence (RFU). EC50 was determined at 23.1 µM.

    4-ONE is a lipid peroxidation product that acts as a TRPA1 (transient receptor potential ankyrin 1) channel activator and displays an EC50 value of 1.9 µM1. The compound contains an electrophilic unsaturated carbonyl moiety and forms adducts with lysine, histidine, and cysteine residues1. 4-ONE is rich in β-sheet structures and is considered to be a reactive aldehyde that may be also cytotoxic2.

    A study conducted with 4-ONE showed that the compound is able to trigger unilateral mechanical hyperalgesia and vasodilatation in mice and also triggersTRPA1-independent effects that relate to edema formation and pain3.

    TRPA1 is a non-selective cation channel expressed in the small neurons of the dorsal root and trigeminal ganglia. TRPA1 channels play an important role in models of inflammatory and neuropathic pain and have the ability to detect pungent or irritant compounds4.
