Peptide (C)DEALKYLSRHHVD, corresponding to amino acid residues 981-993 of rat SERCA3 (Accession P18596). Cytoplasmic, C-terminus.
Western blot analysis of mouse (lanes 1 and 3) and rat (lanes 2 and 4) brain membranes:1,2. Anti-SERCA3 Antibody (#ACP-014), (1:200).
3,4. Anti-SERCA3 Antibody, preincubated with the control peptide antigen.
Expression of SERCA3 in rat cerebellumImmunohistochemical staining of perfusion-fixed frozen rat brain sections with Anti-SERCA3 Antibody (#ACP-014), (1:200), followed by goat anti-rabbit-AlexaFluor-488. SERCA3 immunoreactivity (green) appears in in the molecular layer (MOL) and Purkinje cells (arrows) but not in the granule layer (GL) or cerebellum sections. Cell nuclei are stained with DAPI (blue).
SERCA3 belongs to the family of P-type ion translocating ATPases and is responsible for maintaining a Ca2+ concentration gradient between the cytosol and the endoplasmic reticulum. SERCA3 transports two Ca2+ ions in the endoplasmic reticulum (ER) per ATP molecule hydrolyzed1-3. SERCA proteins are transmembrane and consist of a single polypeptide chain folded into four major domains. SERCA proteins are encoded by 3 genes, ATP2A1, ATP2A2, and ATP2A3. All three genes produce several alternate transcripts and protein isoforms.
SERCA3 protein is mostly expressed in non-muscle cells, especially in endothelial and hematopoietic cells. Minor expression can be detected in muscle cells2,3.
SERCA3 knockout mice exhibit prolonged tail bleeding time and rebleeding1. Mutations in the SERCA3 gene are associated with type 2 diabetes. The protein is also implicated in several cardiac disorders2,3.

Anti-SERCA3 Antibody (#ACP-014) is a highly specific antibody directed against an epitope of the rat protein. The antibody can be used in western blot and immunohistochemistry applications. It has been designed to recognize SERCA3 from rat, mouse, and human samples.
