Origin
Rabbit CaV1.2 cDNA (M57974.1) was transcribed in vitro to produce full length mRNA. CaV1.2 mRNA was injected into Xenopus oocytes and protein expression was verified by detecting voltage dependent Nifedipine (#N-120) sensitive ionic current. Membrane fraction was prepared from oocyte population homogenate and lyophilized. Non-injected oocytes underwent the same procedure excluding mRNA injection.

Activity
(±)-Bay K8644 is a dihydropyridine that acts as an L-type, voltage-gated Ca2+ channel agonist. The (-)-enantiomer has strong Ca2+ channel agonist properties, whereas the (+)-enantiomer has weak Ca2+ channel antagonist activity. The net activity of the racemic mix is L-type channel opening. In the presence of this agonist, channels tend to open for longer periods causing a large increase in the channel microscopic response1-4. (±)-Bay K8644 is also an inhibitor (by increasing the inactivation) of a Shaker KV channel5.
Storage before reconstitution
Shipped at room temperature. Product as supplied can be stored intact at room temperature for several weeks. For longer periods, it should be stored at -20°C.
Reconstitution
Ethanol, methanol or DMSO. Centrifuge all product preparations before use (10000 x g 5 min).
Storage after reconstitution
Up to four weeks at 4°C or three months at -20°C.

Activity
Isradipine is a dihydropyridine that acts as an L-type, voltage-gated Ca2+ channel blocker and is used in the clinic to treat hypertension1,2. Isradipine inhibits L-type channels expressed in Xenopus oocytes with IC50 = 0.2 μM at a holding potential of -80 mV and the inhibition is state dependent and increases with reduced membrane hyperpolarization2. Isradipine, was also shown recently to putatively be an effective treatment for Parkinson’s disease, by affecting particularly the excitability of certain CaV1.3 expressing dopaminergic neurons3.

Reconstitution

Add deionized water, depending on the sample size.

Storage after reconstitution

The reconstituted solution can be stored at 4°C for up to 2 weeks. For longer periods, small aliquots should be stored at -20°C or below.
Avoid multiple freezing and thawing. The further dilutions should be made using a carrier protein such as BSA (1%). Centrifuge all antibody preparations before use (10000 × g 5 min).

Control antigen storage before reconstitution

Lyophilized powder can be stored intact at room temperature for several weeks. For longer periods, it should be stored at -20°C.

Control antigen reconstitution

100 μl water.

Control antigen storage after reconstitution
