Overview
Cat #: BLP-GT001
Type: Synthetic peptide
Form: Lyophilized powder
GABA Transporter 1/GAT-1 (extracellular) Blocking Peptide (#BLP-GT001) is the original antigen used for immunization during Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody (#AGT-001) generation. The blocking peptide binds and ‘blocks’ Anti-GABA Transporter 1/GAT-1 (extracellular) primary antibody, this makes it a good negative reagent control to help confirm antibody specificity in western blot and immunohistochemistry applications. This control is also often called a pre-adsorption control.
Applications: wb, ihc
Application key:
WB- Western blot, IHC- Immunohistochemistry
For research purposes only. not for human use
Applications
Demonstration of Pre-adsorption control
- Western blot analysis of rat (lanes 1 and 3) and mouse (lanes 2 and 4) brain lysates:1,2. Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody (#AGT-001), (1:200).
3,4. Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody, preincubated with GABA Transporter 1/GAT-1 (extracellular) Blocking Peptide (#BLP-GT001). - Expression of GABATR in rat cerebellumImmunohistochemical staining of rat cerebellum using Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody (#AGT-001), (1:200). A. GAT-1 staining is shown in green. B. Neurons expressing gamma amino butyric acid (GABA) were labeled with mouse Anti-parvalbumin (red). C. Merge of the two images demonstrates partial colocalization (arrows). GAT-1 appears both in the soma of Purkinje cells (vertical arrows) and in the pinceau (horizontal arrows).
- Western blot analysis of rat brain lysates (lanes 1 and 3) and mouse brain lysate (lanes 2 and 4):1-2. Guinea Pig Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody (#AGT-001-GP), (1:200).
3-4. Guinea Pig Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody, preincubated with GABA Transporter 1 (GAT-1) (extracellular) Blocking Peptide (#BLP-GT001). - Expression of GABA Transporter 1 in mouse cerebellum.Immunohistochemical staining of perfusion-fixed frozen mouse brain sections with Guinea Pig Anti-GABA Transporter 1 (GAT-1) (extracellular) Antibody (#AGT-001-GP), (1:300), followed by goat anti-guinea pig-AlexaFluor-594. A. GAT-1 immunoreactivity (red) appears in pinceau formations (arrows) around Purkinje cells (P). B. Pre-incubation of the antibody with GABA Transporter 1 (GAT-1) (extracellular) Blocking Peptide (#BLP-GT001), suppressed staining. Cell nuclei are stained with DAPI (blue). M = molecular layer, P = Purkinje cell layer.
